Hexokinase type II (HK2) is an enzyme that catalyzes the first committed step in glycolysis: the phosphorylation of glucose to glucose-6-phosphate. It is the predominant hexokinase isoform in skeletal muscle and plays a crucial role in cellular energy metabolism. HK2 is overexpressed in many cancers, contributing to the Warburg effect. It also has non-metabolic functions, including modulation of transcriptional networks. HK2 localizes primarily to the outer mitochondrial membrane. Expression of HK2 is regulated by insulin, hypoxia, and other factors.
