The hexosamine biosynthesis pathway (HBP) is a metabolic route that integrates glucose, glutamine, acetyl-CoA, and uridine triphosphate (UTP) to produce uridine diphosphate N-acetylglucosamine (UDP-GlcNAc). UDP-GlcNAc is an essential metabolite used for both N-linked and O-linked glycosylation of proteins—a process critical for modulating protein activity, stability, localization, and expression. Altered HBP flux has been implicated in various diseases including diabetes, cardiovascular disease, and cancer.
