Overview

HHIP-like protein 1 (HHIPL1) is a secreted protein encoded by the *HHIPL1* gene on chromosome 14q32[1][3][5]. It is homologous to the Hedgehog-interacting protein (HHIP) and contains a scavenger receptor cysteine-rich (SRCR) domain and a signal peptide for secretion[3][5]. HHIPL1 enhances Hedgehog signaling, positively regulating smooth muscle cell proliferation and migration, and is pro-atherogenic—meaning its activity promotes the development of atherosclerosis[1]. Genome-wide association studies have identified *HHIPL1* variants as risk loci for coronary artery disease[1][3]. The gene is also hypermethylated in certain adenomas, making it a potential biomarker for these tumors[3]. Unlike its paralog HHIP, no drugs are currently known to target HHIPL1, nor is there evidence that it serves as a direct therapeutic target at this time[3][5]. Further functional characterization and potential for druggability remain to be established.
