Overview

HIG1 hypoxia inducible domain family member 1B (HIGD1B) encodes a protein localized primarily to the cell membrane and is a member of the hypoxia inducible gene 1 (HIG1) domain family. The protein plays a significant role in mediating cellular responses to low-oxygen (hypoxic) and pathological conditions. In cardiomyocytes, HIGD1B helps maintain mitochondrial integrity by interacting with optic atrophy 1 (OPA1), which delays its cleavage, stabilizes mitochondrial structure, and prevents apoptosis via inhibition of caspase activation. Aberrant expression of HIGD1B has been observed in various tumor types, particularly in different forms of pituitary adenomas and in lung adenocarcinoma, suggesting a role in tumorigenesis, tumor progression, and potentially as a biomarker or druggable target for these cancers. However, no specific drugs or direct mechanisms of pharmacological action have been described for HIGD1B as of current literature. The protein structure and gene details have been characterized, but it is not classified as a typical receptor or enzyme; its functions relate primarily to hypoxia adaptation and mitochondrial biology rather than classic receptor-ligand signaling or enzymatic activity.
