High-affinity orthophosphate transporter-like protein 1 (HOL1) is a member of the Major Facilitator Superfamily (MFS) and the Cation-polyamine transporter (CPT) family, primarily identified in Saccharomyces cerevisiae [1, 2]. It functions as a multi-specific transporter capable of importing polyamines such as spermidine and putrescine, which are vital for cellular processes like DNA stability and protein synthesis [3, 4]. HOL1 also mediates the transport of various cations and xenobiotics, including the herbicide paraquat [1, 3]. Due to the essential nature of polyamines for fungal growth, HOL1 is investigated as a potential target for antifungal agents that aim to disrupt polyamine homeostasis [4]. Additionally, it serves as a fundamental model for studying polyamine transport mechanisms, which are often exploited by cancer cells to maintain high proliferative rates [3, 4].
