HMGA1P8 (HMGA1-p) is one of eight processed pseudogenes of HMGA1 in the human genome. While it does not encode a protein, HMGA1-p has emerged as a functional RNA that can regulate gene expression through competitive binding to RNA stability factors (specifically αCP1), thereby destabilizing HMGA1 mRNA. Overexpression of HMGA1-p has been shown to decrease HMGA1 protein levels, leading to reduced expression of insulin receptor (INSR) and contributing mechanistically to insulin resistance and type 2 diabetes mellitus. There is also evidence of its involvement in cancer via epigenetic and miRNA-mediated regulatory pathways. Unlike canonical therapeutic targets, HMGA1-p operates chiefly through RNA-mediated gene regulation and is not targetable by traditional pharmacological agents.
