Overview

High mobility group box 1 pseudogene 44 (HMGB1P44) is a pseudogene related to the high mobility group box 1 (HMGB1) gene. Unlike HMGB1, which codes for a chromatin-binding protein involved in DNA architecture and stress responses, HMGB1P44 does not encode a functional protein. Pseudogenes are generally thought to arise from duplication or retrotransposition events and frequently acquire mutations that prevent them from producing functional proteins. While some pseudogenes can regulate their parental genes at the RNA level through mechanisms such as ceRNA activity, there is currently no evidence that HMGB1P44 has any functional regulatory or disease role. It is not considered a therapeutic target, and no drugs or known mechanisms of action are associated with it. The designation "high-mobility group box 2 pseudogene" is not widely supported in curated gene nomenclature databases and may be incorrect. HMGB1P44 should not be confused with HMGB1 (High mobility group box 1 protein), an active protein with nuclear and extracellular functions, or HMGB2, another nuclear protein. Both HMGB1 and HMGB2 are part of the HMGB protein family, but HMGB1P44 does not encode either protein.
