Overview

High mobility group box 1 pseudogene 7 (HMGB1P7) is a processed pseudogene that does not code for a protein. Unlike the parental high mobility group box 1 protein (HMGB1), which has nuclear, cytoplasmic, and extracellular functions as a DNA-binding protein and damage-associated molecular pattern (DAMP), HMGB1P7 exerts indirect biological effects by acting as a competing endogenous RNA. It is reported to sequester miRNAs that would otherwise target HMGB1 mRNA, thereby upregulating HMGB1 expression. Such ceRNA activity has been implicated in the development and progression of some cancers, including breast and endometrial carcinoma, through modulation of cell proliferation and tumorigenesis. HMGB1P7 is not considered a conventional molecular target such as a receptor or enzyme, and there are no drugs that specifically target this pseudogene.

Biological functions

Competes as a competing endogenous RNA (ceRNA) for miRNA binding, indirectly regulating its parental gene HMGB1 expression in some cancers

Disease associations

Cancer (has been reported to act as an oncogenic ceRNA promoting parental gene upregulation in some tumor types)Other (role is via RNA-based regulation, not protein function)
