Overview

High mobility group box protein 1 (HMGB1) is a highly conserved, non-histone chromatin-associated protein with diverse roles in cellular biology and disease. It acts as an architectural chromatin-binding factor, facilitates transcription, and interacts with nucleosomes. Extracellularly, it functions as a damage-associated molecular pattern molecule (DAMP), binding to TLRs and RAGE to trigger pro-inflammatory signaling. HMGB1's compartment-specific functions make it central to both normal cellular processes like gene expression regulation and pathological processes involving inflammation or cancer progression.
