Overview

High mobility group nucleosomal binding domain 2 (HMGN2) is a multifunctional, non-histone chromosomal protein of the high mobility group (HMG) family that binds specifically to nucleosomal DNA and is broadly associated with transcriptionally active chromatin[2][3]. HMGN2 regulates chromatin architecture by modulating nucleosome positioning and counteracting ATP-dependent chromatin remodeling complexes, which fine-tunes gene expression. It binds to both the acidic patch on the histone H2A-H2B dimer and to nucleosomal DNA, stapling the histone core to the DNA and affecting accessibility to regulatory factors and histone H1[1]. HMGN2 is dynamically regulated by post-translational modifications, including acetylation and phosphorylation, which influence its binding to chromatin and its role in transcriptional regulation[2]. Beyond regulation of gene expression, HMGN2 is secreted by activated leukocytes and T-cells, exhibiting direct antimicrobial properties and contributing to tumor cell apoptosis and modulation of host immune responses[2][3]. HMGN2 also impacts autophagic flux in response to cell stress, and its deficiency has been linked to neurodevelopmental defects[2]. There are currently no known drugs directly targeting HMGN2.
