Overview

High mobility group nucleosome-binding domain-containing protein 5 (HMGN5) is a member of the HMGN protein family, characterized by its ability to specifically bind nucleosomes, unfold chromatin, and modulate the transcriptional profile of cells. HMGN5 is localized mainly to euchromatin regions, where it counteracts chromatin compaction mediated by linker histones such as H1, promoting a less condensed chromatin structure. The protein contains an intrinsically disordered, highly acidic C-terminal domain, which distinguishes it from other HMGN variants and enables interactions that influence chromatin architecture. HMGN5 is ubiquitously expressed at low levels across vertebrate tissues, and although it modulates expression of thousands of genes, its individual effects are typically modest. It is not considered a classical therapeutic drug target but represents a chromatin architectural regulator with possible relevance in differentiation, hepatic metabolism, and, potentially, cancer biology[1][2][3][4].
