Overview

High mobility group protein 20A (HMG20A) is a chromatin regulator containing a DNA-binding HMG box, a coiled-coil domain for protein–protein interactions, and an intrinsically disordered N-terminal transactivation domain[2][4]. It primarily localizes to the nucleus and binds non-sequence-specific DNA, modulating chromatin accessibility and influencing gene expression[1][2][3][4]. HMG20A antagonizes the LSD1/CoREST chromatin silencing complex, promoting neuronal differentiation and activating neuronal-specific genes[1][2]. The protein interacts with PHF14 and other chromatin remodeling complexes including NuRD[4], forming multiprotein regulatory structures that affect cell migration, invasion, cell adhesion, and epithelial–mesenchymal transitions (EMT)[2][4]. HMG20A is implicated in several physiological and pathological processes, including type 2 and gestational diabetes (by modulating pancreatic islet beta cell function), neurodevelopment, cancer cell migration and EMT, and possibly tissue fibrosis[1][2][4]. It does not currently have direct clinically approved drugs targeting it, nor is it considered a primary therapeutic target, but it could be relevant in biomarker discovery and future intervention strategies for related diseases[1][2].
