Overview

High mobility group box 2 (HMGB2) is a highly conserved, ubiquitously expressed non-histone chromosomal protein encoded by the HMGB2 gene. It belongs to the high-mobility group (HMG) superfamily, specifically the HMGB subfamily, which is characterized by two DNA-binding HMG-box domains. HMGB2 acts as a chromatin architectural factor that bends and loops DNA, facilitating the assembly of nucleoprotein complexes, modulating chromatin structure, and regulating gene transcription. Beyond architectural functions, HMGB2 is implicated in cellular processes such as response to DNA damage, cell proliferation, differentiation, and aging. Dysregulation of HMGB2 has been linked to cancer, autoimmunity, inflammation, and cellular senescence. While therapeutically attractive due to its central regulatory role, direct targeting of HMGB2 presents significant safety and selectivity challenges due to its widespread nuclear functions
