Overview

HTRA1 is a secreted serine protease enzyme involved in extracellular matrix remodeling, TGF-beta regulation, and protein quality control. It functions as a homotrimer and its dysregulation is implicated in CARASIL, age-related macular degeneration, osteoarthritis, Alzheimer’s disease, and cancer. It presents both potential benefits and challenges as a therapeutic target. Inhibiting HTRA1 might promote TGF-beta signaling, which could have variable effects depending on the context (e.g., promoting fibrosis in some cases). Conversely, enhancing HTRA1 activity could potentially be beneficial in Alzheimer's by promoting tau aggregate degradation but may disrupt normal tissue remodeling. Careful consideration of tissue-specific effects and potential off-target activities would be essential in developing HTRA1-targeted therapies.
