The Hippo-YAP pathway is a highly conserved signaling cascade that regulates organ size, cell proliferation, apoptosis, and tissue homeostasis. It involves a core set of kinases (MST1/2, LATS1/2), scaffold proteins (SAV1, MOB1A/B), and the transcriptional coactivators YAP and TAZ, which bind to TEAD transcription factors. When the pathway is active, YAP/TAZ are phosphorylated and sequestered in the cytoplasm or degraded, inhibiting gene transcription. When the pathway is inactive, YAP/TAZ translocate to the nucleus and promote gene expression, leading to cell growth and survival. Dysregulation of the Hippo-YAP pathway is implicated in cancer and other diseases.
