Overview

Histatin-1 is a small, histidine-rich, cationic peptide primarily found in human saliva, encoded by the *HTN1* gene. It is a key component of the oral innate immune system, acting as an antimicrobial and antifungal agent against pathogens such as *Candida albicans*. Histatin-1 also plays crucial roles in wound healing, stimulating cell adhesion, spreading, and migration—processes essential for tissue repair and angiogenesis in the oral mucosa. The peptide is capable of being internalized by host cells, where it enhances mitochondrial metabolism and activates multiple signaling pathways (such as ERK1/2 and Rac1 GTPase) to promote cell migration and wound closure. In addition, phosphorylation at serine residue 2 increases its affinity for binding to hydroxyapatite on tooth enamel, contributing to the formation of the protective enamel pellicle. Although it has no established role as a pharmacologic or drug target, Histatin-1 and related histatins are studied for their potential in therapeutic and dental applications due to their broad antimicrobial and wound-healing properties[1][2][3][4][5].
