Histidine decarboxylase (HDC) is the enzyme responsible for catalyzing the decarboxylation of L-histidine to histamine. It is a pyridoxal phosphate (PLP)-dependent enzyme and functions as a homodimer. HDC is highly expressed in mast cells, basophils, and certain myeloid cells, and is inducible at sites of inflammation. Its suppression can reduce histamine production, thereby modulating processes dependent on histamine signaling such as inflammation, immune responses, and neurotransmission. Unlike antihistamines that block downstream receptors, HDC suppression addresses both stored and de novo histamine synthesis pathways.
