Overview

Histidine triad nucleotide-binding protein 2 (HINT2) is a mitochondrial enzyme belonging to the histidine triad hydrolase superfamily, mainly expressed in the liver and pancreas[1]. It hydrolyzes nucleotide phosphoramidates, such as adenosine 5’-monophosphoramidate, and participates in mitochondrial regulation of apoptosis as well as steroidogenesis in specific tissues[1][3][5]. HINT2 acts as a tumor suppressor and its down-regulation has been associated with hepatocellular carcinoma and relates to poorer patient prognosis[1][2]. It is not currently known to be a direct drug target and no specific therapies are designed against it, but its expression and enzymatic activity implicate it in mitochondrial health, cell death sensitivity, and potentially in metabolic and proliferative diseases[1][3][5].
