Overview

H2-IAd is a mouse Major Histocompatibility Complex (MHC) class II molecule of the d haplotype, typically found in mouse strains such as BALB/c (The Jackson Laboratory, 2023). It is a cell-surface heterodimer consisting of an alpha chain (H2-Aa) and a beta chain (H2-Ab1) that functions as a critical mediator of the adaptive immune system (UniProt P04224, P06340). The molecule's primary role is to capture peptides derived from extracellular proteins and present them to CD4+ T-cell receptors, which is essential for the activation of T helper cells and subsequent B-cell antibody production (Janeway et al., 2001). In research, H2-IAd is a focal point for studying immunological tolerance, vaccine efficacy, and the mechanisms of autoimmune disorders (PubMed: 15634887). While it is a murine protein, it serves as a vital surrogate for human HLA class II molecules in preclinical drug discovery and mechanistic studies of immune-mediated diseases. Therapeutic strategies involving this target often utilize monoclonal antibodies like MK-D6 to modulate immune responses or specific peptide ligands to induce antigen-specific tolerance (PubMed: 3085232).
