Histone deacetylase 1 (HDAC1) mRNA is the transcript that encodes the HDAC1 enzyme, a critical member of the Class I histone deacetylase family. The resulting protein plays a central role in epigenetic regulation by removing acetyl groups from lysine residues on histones H3 and H4, leading to a condensed chromatin state and transcriptional silencing (UniProt P50486). HDAC1 is frequently overexpressed in various malignancies, where it contributes to the silencing of tumor suppressor genes, promotes cell proliferation, and inhibits apoptosis (PubMed: 22429471). While traditional therapeutics target the HDAC1 protein using small-molecule inhibitors, targeting the mRNA directly via RNA interference (RNAi) or antisense oligonucleotides (ASOs) offers a strategy to achieve higher isoform specificity and reduce the overall protein pool. This approach is particularly relevant in oncology and neurology, where precise modulation of the epigenetic landscape is required to restore normal cellular homeostasis (PubMed: 30554174).
