Histone deacetylase 4 (HDAC4) is an enzyme that removes acetyl groups from lysine residues on histones, leading to chromatin condensation and transcriptional repression. It belongs to the class IIa family of histone deacetylases. HDAC4 plays a crucial role in regulating gene expression involved in heart development, skeletal muscle development, bone formation, neuronal survival, inflammatory responses, and metabolic pathways. Its activity is regulated by post-translational modifications, including phosphorylation, which controls its subcellular localization. Alterations in HDAC4 are implicated in various diseases, including intellectual disability syndromes, metabolic/inflammatory diseases, and cancer.
