Overview

Histone H1.0 is a linker histone variant predominantly found in differentiated cells and is involved in packaging DNA into higher-order chromatin structures by binding to the linker DNA between nucleosomes[1][2][3][4]. It plays an essential role in chromatin compaction, nucleosome spacing, and regulation of gene expression by influencing genome accessibility. H1.0 is structurally characterized by an unstructured N-terminal domain, a central globular domain (containing a winged-helix DNA binding motif), and an unstructured, highly basic C-terminal domain[2]. Recent studies indicate that H1.0 is necessary and sufficient for fibroblast activation, affecting cellular mechanics, gene expression (notably of extracellular matrix and cytoskeletal genes), and fibrosis development[3]. While post-translational modifications modulate its function, and its levels are implicated in both normal and disease states (especially in cancer and fibrotic diseases), there are currently no clinically used drugs targeting H1.0 directly. H1.0 is not considered a classic druggable target (such as a receptor or enzyme), but its role as a biomarker for cell differentiation and disease is under investigation[1][2][3].
