Histone H2A-like 3 (H2A.L.3) is a member of the histone H2A variant family, which forms part of the octameric core of the nucleosome, the fundamental organizational unit of chromatin in eukaryotic cells[3][5]. Histone variants, including H2A.L.3, contribute to chromatin diversity by replacing canonical H2A in nucleosomes, thereby influencing nucleosome stability, higher-order chromatin structure, and chromatin function. Although core histones are highly conserved, H2A variants differ mostly in their C-terminal regions and may confer specific structural and functional properties to nucleosomes, such as altering DNA accessibility and interactions with other chromatin factors[3]. H2A variants are critical in developmental, tissue-specific, or stress-responsive chromatin states, but H2A.L.3 itself is not a validated therapeutic target, nor are there approved drugs or standard biomarker roles associated with it at present[3][4][5].
