Overview

Histone H2B type 1-A is a basic nuclear protein essential for the nucleosome structure of chromatin in eukaryotes. It is a core component of the histone octamer around which DNA is wrapped, facilitating DNA packaging and higher order chromatin structure. This variant is specifically expressed in testis and sperm cells, where it plays a critical role in replacing conventional H2B histones, helping to remodel chromatin and enable the eventual transition from nucleosomes to protamine during spermatogenesis. The gene encoding this protein is intronless and produces a replication-dependent transcript that contains a palindromic termination element. Besides its well-known role in chromatin compaction and regulation, H2B type 1-A is involved in epigenetic modification and downstream events in male germ cells. Its dysregulation or mutation is associated with male infertility and may have secondary implications for chromatin-related diseases[1][3][4][5][7]. Note: Histone H2B type 1-A (H2BC1) is not considered a therapeutic target like receptors, enzymes, or transporters commonly targeted by drugs. Its roles are mainly structural and regulatory within chromatin, particularly in testis biology.
