Histone H2B type 1-H (H2BC9) is a member of the core histone H2B family and is a basic nuclear protein crucial for establishing the nucleosome structure in eukaryotic chromosomes[1][2][9]. Two molecules each of H2B, H2A, H3, and H4 form an octamer at the core of the nucleosome—about 146 base pairs of DNA wrap around this structural unit, compacting DNA into chromatin and regulating its accessibility. H2BC9 is encoded by an intronless gene located in the major histone gene cluster on human chromosome 6. This protein is replication-dependent and functions in DNA packaging, transcriptional regulation, DNA repair, DNA replication, and chromosomal stability, in part through participating in post-translational modifications that constitute the histone code[1][2][9]. There is no evidence that it is a direct therapeutic target or that specific drugs act on it; however, alterations in histone expression and post-translational modification are implicated in various diseases, most notably cancer[1][7].
