Histone H2B type 3-B (H2BC26) is a replication-dependent nuclear protein that forms one of the four core histones responsible for the nucleosome structure in eukaryotic cells. The nucleosome consists of DNA wrapped around an octamer of histones (two molecules each of H2A, H2B, H3, and H4), providing compaction and organization to chromatin. H2BC26, specifically, is an isoform of H2B, and contributes to core chromatin functions including transcription regulation, DNA replication, DNA repair, and maintenance of genome stability through post-translational modifications such as acetylation, phosphorylation, and ubiquitination. While H2BC26 is fundamentally structural and not typically considered a druggable therapeutic target, its altered expression or modification has been associated with certain diseases such as hematologic cancer. There is no evidence for small-molecule drugs or biological agents directly targeting this specific isoform, and interfering with core histones generally poses significant safety concerns due to their essential role in cell viability.
