Overview

Conserved regions in HIV-1 refer to stretches of the viral genome or protein sequences that remain relatively unchanged across different viral strains and subtypes. These regions are critical for essential viral functions, such as entry, replication, and assembly, making them attractive targets for therapeutic interventions and vaccine design. Key conserved regions include the CD4 binding site (CD4bs) and V3 loop on gp120, N-heptad repeat (NHR), fusion peptide proximal region (FPPR), membrane-proximal external region (MPER), and fusion peptide (FP) on gp41, and core domains of reverse transcriptase (RT). These regions are targeted by antiretroviral drugs and are the focus of vaccine development efforts to elicit broadly neutralizing antibodies and T cell responses.
