Overview

The HIV-1 envelope glycoprotein (Env) is a trimeric complex of gp120 and gp41 subunits on the surface of HIV-1 virions. It mediates viral entry into host cells by binding to CD4 and a coreceptor (CCR5 or CXCR4), triggering membrane fusion. Env is the primary target for neutralizing antibodies and vaccine development, but its high variability and glycosylation pose significant challenges.

Mechanism of action

Entry inhibitors block viral entry by targeting gp41 (enfuvirtide) or CCR5 (maraviroc). bNAbs block viral entry by binding to conserved epitopes on gp120 and/or gp41.
