The membrane fusion process mediated by the HIV-1 envelope glycoprotein (Env) complex is essential for viral entry into host cells. This process involves the interaction of gp120 with CD4 and a coreceptor (CCR5 or CXCR4), leading to conformational changes in gp41 and insertion of the fusion peptide into the target cell membrane. The subsequent formation of a six-helix bundle brings the viral and cellular membranes together, facilitating fusion. This process can be inhibited by peptides like C46, which block the formation of the six-helix bundle.
C46 peptide binds to the HR1 region of gp41, preventing the formation of the six-helix bundle and inhibiting membrane fusion.
