Overview

This target represents a cell-based immunotherapeutic complex consisting of alpha-type-1 polarized dendritic cells (αDC1) loaded with conserved peptides derived from the HIV-1 Gag and Pol proteins. αDC1 are specialized antigen-presenting cells generated in vitro using a specific maturation cocktail (e.g., IL-1β, TNF-α, IFN-α, IFN-γ, and poly-I:C) that enables them to produce high levels of IL-12p70, which is essential for inducing potent Th1 and cytotoxic T lymphocyte (CTL) responses (Mailliard et al., 2004, PMID: 15208254). The peptides used are selected from highly conserved regions of the viral proteome to minimize the risk of viral mutational escape, a major challenge in HIV treatment (Macatangay et al., 2016, PMID: 27163323). These peptides are presented on the surface of the αDC1 via Major Histocompatibility Complex (MHC) Class I and II molecules to prime and expand HIV-specific T cells in the patient. This therapeutic approach aims to enhance the immune system's ability to recognize and eliminate HIV-infected cells, particularly those in the latent reservoir. Clinical evaluation of this target focuses on its ability to maintain viral suppression and reduce the need for lifelong antiretroviral therapy (Rinaldo et al., 2014, PMID: 24403414).
