Overview

HIV-1 Gag-derived p24 and Pol-derived RT epitopes are peptide fragments obtained from the HIV-1 capsid protein (p24, encoded by the gag gene) and the HIV-1 reverse transcriptase enzyme (encoded by the pol gene). These epitopes are used extensively in immunological research, especially to study virus-specific T cell responses in infected individuals or in vaccine trials, due to their conservation among HIV-1 variants and their importance in the viral life cycle. While p24 and RT are the targets of drug intervention and play structural and enzymatic roles within the virus, their epitopes serve as dominant targets for T cell-mediated immunity. However, the term as used here does not refer to a specific, single molecular target but rather to a collection of antigenic peptides utilized for immune monitoring and vaccine development.

Mechanism of action

Not applicable to the epitopes themselves. As epitopes: They are recognized by cytotoxic T lymphocytes (CTLs) during immune surveillance, with their presentation on MHC class I molecules leading to CTL activation and potential killing of infected cells.

Safety considerations

None unique to the epitopes themselves (General vaccine/immune manipulation concerns may apply if used in immunotherapy contexts).

Biomarkers

Used as biomarkers for cellular immune response to HIV-1, e.g., measuring HIV-specific T cell function in clinical trials and diagnostics
