HIV-1 p24-RT-Nef-p17 is a synthetic multi-antigen fusion protein designed for use in HIV-1 vaccines and immunotherapies. It combines four key proteins from the Human Immunodeficiency Virus type 1: the capsid protein (p24), reverse transcriptase (RT), the negative regulatory factor (Nef), and the matrix protein (p17) (García et al., 2011, The Lancet Infectious Diseases). These components are selected to provide a broad range of T-cell epitopes to stimulate a robust and diverse immune response against the virus, particularly targeting conserved regions to minimize the impact of viral mutations (Gómez et al., 2011, Vaccine). In clinical development, this construct is often delivered via viral vectors like Modified Vaccinia Virus Ankara (MVA-B) or as DNA vaccines (DNA-B). The goal of targeting these proteins is to control viral replication and reduce the viral reservoir in infected individuals, potentially leading to a functional cure or improved management of HIV/AIDS by enhancing the host's cellular immunity (Gómez et al., 2012, Journal of Virology). The fusion protein acts as a comprehensive immunogen that allows the immune system to recognize multiple stages of the viral life cycle, from entry and reverse transcription to assembly and release.
