Overview

The NNRTI binding site is a hydrophobic pocket on the p66 subunit of HIV-1 reverse transcriptase. It is the target for non-nucleoside reverse transcriptase inhibitors (NNRTIs), which are allosteric inhibitors that bind to this site and induce conformational changes in RT, thereby inhibiting its enzymatic activity and preventing viral replication. The plasticity of this pocket and the emergence of resistance mutations are major challenges in HIV therapy.
