The HIV Gag peptide–HLA-E complex is formed when the nonclassical class Ib MHC molecule HLA-E presents a peptide derived from the HIV-1 Gag protein, such as RL9 (RMYSPTSIL) or KF11 (KRWIILGLNK), on the surface of infected cells[1][3][4][5]. Unlike most HLA-E complexes that display highly conserved leader peptides to regulate NK cell activity by engaging CD94/NKG2 receptors, the presentation of HIV-specific peptides allows for recognition by a unique subset of HLA-E–restricted CD8+ T cells that can suppress HIV-1 replication[3][4][6]. These T cell responses are vaccine-inducible in vitro and have been shown to inhibit viral replication[3][6]. The complex represents a potential immunotherapeutic and vaccine target in HIV infection, providing an alternative immune attack strategy distinct from classical HLA class I antigen presentation, but with unique regulatory challenges and implications for immune modulation[5][6][7].
