Overview

The HLA-A*02:01–survivin (95-104) peptide complex is a specific molecular target consisting of a decameric fragment of the survivin protein (BIRC5) presented by the Human Leukocyte Antigen (HLA) allele A*02:01 (UniProt P50607). Survivin is a member of the inhibitor of apoptosis (IAP) family, which is characteristically overexpressed in a vast majority of human cancers while remaining nearly undetectable in most differentiated normal tissues (PMID: 10667374). The 95-104 epitope (sequence: ELTLGEFLKL) is an immunodominant peptide that, when presented on the cell surface, allows the immune system to distinguish malignant cells from healthy ones (PMID: 15930334). Therapeutic interventions targeting this complex include T-cell receptor (TCR) engineered T-cell therapies and peptide-based vaccines like SurVaxM, which aim to induce a robust CD8+ cytotoxic T-lymphocyte response (NCT02455557). By binding specifically to this pMHC complex, these therapies trigger the release of perforins and granzymes, leading to the selective destruction of survivin-expressing tumor cells. This target is particularly relevant in the treatment of glioblastoma, melanoma, and various solid tumors where survivin drives cell survival and treatment resistance.
