Overview

The HLA-A*11:01 presenting POLE P286R neoantigen peptide is a specific peptide-major histocompatibility complex (pMHC) target found on the surface of cancer cells. The POLE gene encodes the catalytic subunit of DNA polymerase epsilon, which is responsible for leading-strand DNA synthesis. The P286R mutation occurs in the exonuclease domain, impairing proofreading and resulting in an ultramutated tumor phenotype. This specific mutation creates a novel amino acid sequence (neoantigen), such as the peptide LAFDIETTK, which is processed and presented by the HLA-A*11:01 allele. Because this mutation is a recurrent hotspot in endometrial and colorectal cancers, the pMHC complex serves as a highly specific target for T-cell-based immunotherapies, such as TCR-engineered T cells or neoantigen vaccines. Targeting this complex allows the immune system to selectively destroy malignant cells while sparing healthy tissue that lacks the mutation.
