The HLA-A24 presenting MPHOSPH1 278-286 epitope is a peptide-major histocompatibility complex (pMHC) consisting of a 9-amino acid fragment (IYNEYIYDL) derived from the M-phase phosphoprotein 1 (MPHOSPH1, also known as KIF20B) presented by the HLA-A*24:02 molecule (PubMed: 15150592). MPHOSPH1 is a kinesin-related motor protein essential for the completion of cytokinesis during the cell cycle (UniProt: Q96Q89). While its expression is minimal in most normal adult tissues, it is significantly upregulated in various malignancies, including bladder, lung, and esophageal carcinomas, making it a viable tumor-associated antigen (TAA). This specific epitope is highly immunogenic and serves as a target for cancer immunotherapy, particularly in East Asian populations where the HLA-A24 allele is prevalent (PubMed: 17459057). Therapeutic strategies targeting this complex include peptide-based vaccines, such as OTS102, and T-cell receptor (TCR) engineered T-cell therapies designed to trigger a cytotoxic T lymphocyte (CTL) response against tumor cells. Clinical trials have demonstrated that targeting this epitope can elicit specific immune responses and potentially improve survival in patients with advanced cancers.
