Overview

The HLA-A*24:02-URLC10-177 complex is a specific peptide-major histocompatibility complex (pMHC) class I molecule that serves as a target for cancer immunotherapy. URLC10 (Up-Regulated in Lung Cancer 10) is a cancer-testis antigen highly expressed in various malignancies, including non-small cell lung cancer and esophageal cancer, while remaining silent in normal adult tissues except for the testis (PubMed: 19448168). The URLC10-177 epitope (sequence: SYWGVIGHL) is processed and presented by the HLA-A*24:02 allele, which is highly prevalent in East Asian populations (PubMed: 22150169). This complex is recognized by specific T-cell receptors (TCRs) on cytotoxic T lymphocytes (CTLs), triggering an immune response against tumor cells. Therapeutic strategies targeting this complex include peptide-based vaccines and TCR-engineered T-cell therapies designed to induce or provide high-affinity recognition of the pMHC (PubMed: 25100374). Clinical trials have shown that targeting this complex can lead to the induction of specific CTLs and potential clinical benefits in patients with URLC10-positive tumors (PubMed: 21347601).
