In laboratory settings, they are essential for validating the performance of assays such as ELISpot, MHC-peptide binding affinity tests, and flow cytometry-based multimer staining (Janeway et al., 2001). By providing a known baseline of immune reactivity, these peptides allow researchers to normalize data and ensure the specificity of T-cell responses to novel antigens or therapeutic candidates. They do not function as therapeutic targets themselves but are critical tools for the development and quality control of vaccines and immunotherapies. Their use is standard in clinical trials to monitor the baseline immune status of participants and the technical consistency of immunogenicity assays.
