Overview

HLA-DQB1 antisense RNA 1 (HLA-DQB1-AS1) is a long non-coding RNA gene located adjacent to the protein-coding HLA-DQB1 gene in the human major histocompatibility complex (MHC) class II region. HLA-DQB1-AS1 does not code for a protein but exerts regulatory functions by binding to specific RNA-binding proteins, notably ZRANB2, to enhance cell proliferation and inhibit programmed cell death (apoptosis). Its upregulation is implicated in the progression of several cancers, particularly hepatocellular carcinoma, and has also been studied in the context of autoimmune and immune-mediated diseases. HLA-DQB1-AS1 exemplifies a growing class of disease-associated lncRNAs with potential utility as prognostic biomarkers and, potentially, as novel therapeutic targets[2][4][5][6][7].

Mechanism of action

Interacts directly with RNA-binding proteins (ZRANB2) to promote cell proliferation and inhibit apoptosis[2][4][5] May act as a regulatory lncRNA in disease-associated gene expression[6]

Biological functions

Regulation of cell proliferation[2][4][5]Inhibition of apoptosis[2][4][5]Potential regulation of immune-related genes[6]RNA/protein binding (notably ZRANB2 protein)[2][4][5]
04

Disease associations

Cancer (specifically hepatocellular carcinoma; evidence also in lung adenocarcinoma, melanoma)[2][4][5]Autoimmune disease (possible link in celiac disease context)[6]Other
05

Safety considerations

No specific safety concerns or therapeutic challenges are reported; general lncRNA-targeted therapies would require careful specificity to avoid off-target effects[4][5]
06

Biomarkers

Upregulated expression of HLA-DQB1-AS1 may serve as a biomarker for proliferation/apoptosis status in hepatocellular carcinoma and possibly prognosis in other cancers[2][4][5]
