Overview

HMG box transcription factor BBX (BBX) is a member of the HMG-box superfamily of DNA-binding proteins, which contains a single HMG (High Mobility Group) box domain[3][5]. BBX acts as a sequence-specific transcription factor, binding double-stranded DNA and regulating transcription by RNA polymerase II[1][2][5]. It is necessary for proper cell cycle progression from G1 to S phase[4]. BBX is predominantly nuclear and has predicted roles in chromatin organization and gene transcription. In mammalian systems, it functions as a regulatory node in forebrain development and may be involved in neurogenic patterning; its expression is notably high in the cortex and hippocampus, suggesting a role in central nervous system development[2]. Variations in the BBX gene region have been associated with risk for Alzheimer’s disease, indicating a potential role in neurodegeneration[1]. The precise mechanisms, interacting partners, and therapeutic or diagnostic utility of BBX in human disease are still largely unknown, and there are no drugs currently known to directly target BBX[2][4][5].
