Overview

HMMR antisense RNA 1 (HMMR-AS1) is a long noncoding RNA transcribed from the antisense strand adjacent to the HMMR gene, which encodes the hyaluronan-mediated motility receptor. HMMR-AS1 is upregulated in basal-like breast cancer and exerts regulatory control over HMMR gene expression. Knockdown of HMMR-AS1 in BLBC cell lines leads to reduced proliferation and migration, and lower HMMR levels, suggesting it acts as a tumor-promoting regulator. HMMR-AS1 expression is also positively correlated with the oncogenes BRCA1 and BRCA2 and negatively correlated with the tumor suppressor PTEN. These properties make HMMR-AS1 a promising diagnostic biomarker and potential therapeutic target in cancer, especially basal-like breast cancer[1].

Mechanism of action

Gene knockdown: siRNA-mediated suppression of HMMR-AS1 reduces cancer cell proliferation and migration by downregulating HMMR expression[1]

03

Biological functions

Regulation of gene expression (particularly of HMMR/RHAMM)Modulation of cell proliferationModulation of cell migration/invasionRegulation of cancer-related genes (BRCA1, BRCA2, PTEN)
04

Disease associations

Cancer (specifically basal-like breast cancer)Potentially other aggressive cancer subtypes (via regulation of HMMR)
05

Safety considerations

Unknown or unreported (safety implications of therapeutically targeting HMMR-AS1 are not described in current literature[1])
06

Biomarkers

HMMR-AS1 itself may serve as a biomarker for basal-like breast cancer diagnosis and progression monitoring
