Homeobox B5 (HOXB5) mRNA is the transcript of a gene belonging to the Antp homeobox family, encoding a transcription factor that serves as a master regulator of positional identity and organogenesis during embryonic development. It plays a vital role in the formation of the lungs, gut, and enteric nervous system, and in adults, it is a key marker and regulator of long-term hematopoietic stem cell self-renewal. In many human cancers, HOXB5 mRNA is significantly upregulated, acting as an oncogenic driver that promotes cell proliferation, invasion, and the epithelial-mesenchymal transition (EMT). Because the HOXB5 protein is a transcription factor and traditionally difficult to target with small molecules, the mRNA itself is the primary focus for therapeutic intervention using RNA-silencing technologies like siRNA and ASOs. High levels of HOXB5 mRNA are often correlated with poor clinical outcomes and resistance to standard treatments, making it both a promising therapeutic target and a valuable prognostic biomarker.
