Overview

Homeobox protein DLX-4 (DLX4) is a vertebrate transcription factor belonging to the distal-less (Dlx) gene family, characterized by a homeobox DNA-binding domain. It regulates embryonic development processes including craniofacial patterning, nervous system formation, and reproductive organ development. Pathologically, DLX4 is upregulated in various cancers and contributes to oncogenesis by promoting cell proliferation, suppressing apoptosis, maintaining cancer stem cell populations, enhancing metastasis through EMT, and remodeling the tumor microenvironment via immune modulation. High DLX4 expression and methylation status serve as prognostic biomarkers in several cancer types, particularly renal clear cell carcinoma, where it is associated with poor outcome and tumor aggressiveness. As a result, DLX4 is considered an emerging therapeutic target, though no direct DLX4-targeted drugs exist to date, and targeting may pose developmental safety risks based on its essential roles in normal tissue formation.

Mechanism of action

No approved small-molecule or biologic drugs directly target DLX4. Basic research suggests that targeting DLX4 may affect cell proliferation, immune infiltration, and EMT processes, but no drug mechanisms have been established in the clinic.
