Overview

Homeobox protein Hox-A11 (HOXA11) is a sequence-specific DNA-binding transcription factor encoded by the HOXA11 gene in humans[3][5]. It is a member of the HOX family of proteins, characterized by a homeodomain that mediates DNA binding, and plays a fundamental role in embryonic morphogenesis, skeletal system development, and tube branching morphogenesis[2][3]. Expression of HOXA11 is strictly regulated during development, and it is required for proper uterine development and female fertility[3]. Mutations in HOXA11 can lead to limb malformations and bone marrow failure syndromes. In cancer, especially lung adenocarcinoma, HOXA11 is frequently overexpressed and can function as an oncogene, promoting tumor cell migration, adhesion, and participation in key signaling pathways such as focal adhesion and extracellular matrix interactions[2]. HOXA11 is also mechanistically important in protein-protein interactions, such as with Forkhead box O1A (Foxo1a), driven by evolutionarily conserved regions within the protein[1].
