Overview

Homeobox protein Hox-A3 (HOXA3) is a DNA-binding, sequence-specific transcription factor that is a member of the homeobox gene family, located in the A cluster on human chromosome 7. It is crucial for embryonic development, where it orchestrates the spatial and temporal regulation of genes required for the morphogenesis of key organs derived from the pharyngeal region, such as the thymus, parathyroid, and thyroid. In the nervous system, HOXA3 is involved in specifying neuronal identity and segmenting the hindbrain. Its expression is tightly controlled and, in adults, can be re-induced during wound healing to promote tissue remodeling and angiogenesis. Pathogenic variants or knockout of HOXA3 lead to severe developmental defects, and its dysregulation has been associated with certain cancers and congenital organ malformations.
