Overview

Homeobox protein Hox-A4 (HOXA4) is a sequence-specific DNA-binding transcription factor that belongs to the homeobox gene family, clustered on human chromosome 7[1][3]. It regulates the expression of numerous target genes critical for embryonic development, morphogenesis, and cell differentiation, particularly in the context of providing spatial positional identity along the anterior-posterior axis[1][3]. The protein operates predominantly by modulating gene transcription, both activating and repressing genes necessary for the proper development of tissues and organs[2]. In adults, HOXA4 maintains tissue homeostasis and preserves vascular integrity. Aberrations in its expression or epigenetic regulation (such as hypermethylation) are associated with several malignancies (notably leukemia and colorectal cancer), congenital anomalies, and vascular diseases (e.g., abdominal aortic aneurysm)[2][3]. Consequently, HOXA4 is an emerging biomarker for disease prognosis and a potential therapeutic target, though not yet addressed by any approved drug[2][3][5].
