Homeobox protein Hox-B4 (HOXB4) is a member of the Antp homeobox family and functions as a sequence-specific transcription factor involved in embryonic development and cell differentiation[1][2][3][4][5]. It is a key regulator in the maintenance, self-renewal, and expansion of hematopoietic stem and progenitor cells. HOXB4 is included in a cluster of homeobox B genes on chromosome 17 and contains a homeobox DNA-binding domain. Experimental overexpression in human and murine cells enhances stem cell self-renewal, with applications in stem cell therapy and hematopoietic reconstitution[1][3][4][5]. In cancer biology, HOXB4 exhibits context-dependent functions, acting as a tumor suppressor in cervical and breast cancer by repressing proliferation and the Wnt/β-catenin pathway, yet potentially functioning as an oncogene in ovarian cancer and other contexts[2]. Expression levels of HOXB4 can serve as a potential biomarker in several malignancies, reflecting its dual role in tumor suppression and oncogenesis depending on tissue and regulatory environment[2][4].
