Overview

Homeobox protein Hox-B5 (HOXB5) is a sequence-specific transcription factor encoded by the HOXB5 gene located in the homeobox B gene cluster on chromosome 17. It is a member of the Antennapedia (Antp) homeobox family, characterized by a conserved DNA-binding homeobox domain. HOXB5 regulates gene expression in embryonic development, with key roles in lung and gut formation and cell lineage specification. In adults, it marks long-term hematopoietic stem cells and contributes to stem cell identity. Aberrant expression of HOXB5 is associated with several human diseases, notably acute myeloid leukemia and multiple solid tumors, where it can drive proliferation, invasion, and epithelial–mesenchymal transition. It also serves as a prognostic biomarker in certain cancers and is implicated in rare congenital malformations of the lung and gut[1][3][5].
