Overview

Homeobox protein Hox-B6 is a sequence-specific transcription factor encoded by the HOXB6 gene, belonging to the Antennapedia (Antp) homeobox family. It contains a homeobox DNA-binding domain and is part of a cluster of homeobox B genes (chromosome 17). HOXB6 directs cellular fate during development, particularly in blood, lung, skin, and skeletal tissue, by regulating gene transcription and chromatin structure. It is localized in both the nucleus and cytoplasm, suggesting canonical and noncanonical functions. Misexpression or altered localization of HOXB6 is implicated in acute myeloid leukemia (often accelerating the disease when co-expressed with MEIS1), colorectal cancer, and developmental skeletal anomalies. HOXB6 activity relies on DNA binding, not necessarily requiring PBX co-factor interaction, but is modulated through associations with chromatin remodeling proteins such as CREB-binding protein (CBP). Disruption of epigenetic regulators (e.g., Polycomb group proteins) can impact its expression. Therapeutic manipulation of HOXB6 faces challenges due to its fundamental roles in development and hematopoiesis[1][2][3][4][6].
